A new biosurfactant produced by Candida glabrata UCP 1002: characteristics of stability and application in oil recovery BABT

The production of a new biosurfactant by Candida glabrata UCP1002 was studied to evaluate the influence of the concentration of the cotton seed oil, glucose and yeast extract. The dynamics of the growth and surfactant production were showed for all the cultivation conditions studied. The best emulsification of the n-hexadecane, quantified by the emulsifying index was observed in the medium containing 7.5% cotton seed oil, 5% glucose and 0.3% yeast extract. Lipopeptides represent a class of microbial surfactants with increasing scientific, therapeutic and biotechnological interests. The genus Bacillus is a producer of these active compounds, and among them B. subtilis produces surfactin, the most potent biosurfactant known. These compounds can act as antibiotics, antivirals, antitumorals, immunomodulators and enzyme inhibitors. Background: The aim of the present study was to propose a low-cost method for the production of a biosurfactant by the yeast Candida sphaerica and assess its toxicity and phytotoxicity. The medium was formulated with distilled water supplemented with residue from a soy oil refinery (5%) and corn steep liquor (2.5%) as substrates. These two products were the sources of carbon and nitrogen as well as mineral elements to encourage the growth of the microorganism and production of a biosurfactant. The aim of this study was to increase rhamnolipid production by formulating media using kefir and fish meal for Pseudomonas aeruginosa strains isolated from different environmental resources. The strains, named as H1, SY1, and ST1, capable of rhamnolipid production were isolated from soil contaminated with wastes originating from olive and fish oil factories. Additionally, P. aeruginosa ATCC 9027 strain, which is known as rhamnolipid producer, was included in the study. Background: Biotechnological processes are costly, especially for the production of biosurfactants. The successful production of a biosurfactant is dependent on the development of processes using low cost raw materials. Considering the importance of the characteristics of a biosurfactant to facilitate its industrial application, the properties of the biosurfactant produced by Candida lipolytica through previously optimized medium have been established. This study focused on the isolation and characterization of bacterial strains with superior oil degrading potential from crude-oil contaminated soil. Three such bacterial strains were selected and subsequently identified by 16S rRNA gene sequence analysis as Corynebacterium aurimucosum, Acinetobacter baumannii and Microbacterium hydrocarbonoxydans respectively. Crude glycerol from biodiesel industry as substrate for biosurfactant production by Bacillus subtilis ATCC 6633 BABT

Glycerol, a co-product of the biodiesel industry, may be a suitable raw material for the production of high added-value compounds by the microorganisms. This study aimed to use the glycerol obtained from the biodiesel production process as the main carbon source for biosurfactant production by Bacillus subtilis ATCC 6633. Results indicated that the strain lowered the surface tension of the cell-free fermented broth to 31.5 ± 1.6 mN/m, indicating the production of biosurfactant. The critical micelle concentration (CMC = 33.6 mN/m) obtained was similar to the previously reported for biossurfactants isolated from other Bacillus. The produced biosurfactant was able to emulsify n-hexadecane and soybean oil.

A lipopeptide biosurfactant produced by Bacillus natto TK-1 has a strong surface activity. The biosurfactant was found to be an anti-adhesive agent against several bacterial strains, and also showed a broad spectrum of antimicrobial activity. The biosurfactant induced a significant reduction in tumor cells viability in a dose- dependent manner.

Evaluation of emulsifier stability of biosurfactant produced by Saccharomyces lipolytica CCT-0913 BABT

Surface-active compounds of biological origin are widely used for many industries (cosmetic, food, petrochemical). The Saccharomyces lipolytica CCT-0913 was able to grow and produce a biosurfactant on 5% (v/v) diesel-oil at pH 5.0 and 32ºC. The cell-free broth emulsified and stabilized the oil-in-water emulsion through a first order kinetics. The results showed that the initial pH value and temperature influenced the emulsifier stability (ES), which was the time when oil was separated. Lipopeptide production by Bacillus subtilis R1 and its possible applications BJM

Abstract The possible application of a bacterial strain - Bacillus subtilis R1, isolated from an oil contaminated desert site in India, as biocontrol agent and its biosurfactant in microbial enhanced oil recovery are discussed. The biosurfactant production in minimal medium was carried out at different temperatures and salt concentrations, where it produced an efficient biosurfactant at 30-45 °C and in presence of up to 7% salt. It significantly reduced the surface tension from 66 ± 1.25 mN/m to 29 ± 0.85 mN/m within 24 h. In order to enhance the biosurfactant production, random mutagenesis of B. subtilis R1 was performed using chemical mutagen - ethyl methanesulfonate. In this work we investigated the structure of the iron-stimulated surface tension reducing substances produced by P. citronellolis 222A isolated from a 17-years old landfarming used for sludge treatment in petrochemical industries and oil refinery. Its mass spectrum differs from P. aeruginosa spectrum, indicating that the surface tension reducing substances produced by P. citronellolis can be a new kind of biosurfactant.

Production and characterization of a biosurfactant produced by Streptomyces sp. DPUA 1559 isolated from lichens of the Amazon region BJMBR

Surfactants are amphipathic compounds containing both hydrophilic and hydrophobic groups, capable to lower the surface or interfacial tension. Considering the advantages of the use of biosurfactants produced by microorganisms, the aim of this paper was to develop and characterize a biosurfactant produced by Streptomyces sp. DPUA1559 isolated from lichens of the Amazon region. The microorganism was cultured in a mineral medium containing 1% residual frying soybean oil as the carbon source. The kinetics of biosurfactant production was accompanied by reducing the surface tension of the culture medium from 60 to values around 27.14 mN/m, and by the emulsification index, which showed the efficiency of the biosurfactant as an emulsifier of hydrophobic compounds....

Production and characterization of surface-active compounds from Gordonia amicalis BABT

Two methods were used to make crude preparations of surface-active compounds (SACs) produced by Gordonia amicalis grown on the medium containing 1% diesel oil. Using a 2:1 (v/v) solution of chloroform:methanol for extraction, Type I SACs were isolated and shown to produce oil in water (O/W) emulsions. Type II SACs were isolated by precipitation with ammonium sulfate and produced predominantly water in oil emulsions (W/O). The crude Type I and II preparations were able to produce a significant reduction in the surface tension of water; however, the crude Type II preparation had 10-25 fold higher emulsification activity than the Type I preparation. Evaluation of both tenso-active and emulsifying activities indicated that a biosurfactant was produced by the newly isolated and promising strain Candida glabrata isolated from mangrove sediments. The extracellular water-soluble emulsifying agent was isolated and identified as a heteropolymer. The maximum of bioemulsifier production was observed when the strain was grown on soluble and insoluble substrates cotton seed oil plus glucose, reaching values of 10.0 g/l after 144 hrs at 200 rpm. The cell-free culture broth containing the examined agent lowered the surface tension of the medium to 31 mN/m. Selection of microorganisms for biosurfactant production using agroindustrial wastes BJM

Biosurfactant production by some bacterial isolates using molasses, milk whey and cassava flour wastewater (manipueira) as substrates was evaluated and compared with the production in conventional medium. Isolates growing in manipueira medium decreased the surface tension around 42%, the highest reduction among all the substrates tested. From the eleven isolates tested, eight were able to decrease the surface tension to levels below 30 mN/m using manipueira as substrate. The isolates LB5a, LB2a, LB262, LBB and LB1a that gave surface tension about 26 mN/m were identified as Bacillus sp.
